伊莉莎白·肖（Elizabeth Shaw）：頂尖科學家；古生物學、考古學、神話學及仿生學博士；科學考察船「普羅米修斯號」（Prometheus）首席科學官。曾在洞窟發現一組原始壁畫，找到創造主造人的有力證據，奮力向彼得·維蘭德推介自己的『工程師（Engineers）創造主』一說，最終成功帶領一眾科學家登上普羅米修斯號，出發前往行星LV-223，開展創造主探究之旅。老公是查理·霍洛威博士。伊莉莎白是大衛所心慕之人。

大衛（David）：維蘭德-尤坦尼集團（Weyland-Yutani）出產的大衛第八型仿生人，普羅米修斯號配備的隨行仿生人，一個超越普通人造意識的仿生機械個體。隨著普羅米修斯號的創造主探究旅程，他漸漸衍生了更為複雜的個體意識；並因伊莉莎白博士在旅程中顯露的堅強個性，而對她產生尊敬與愛慕的人性情感。

查理·霍洛威（Charlie Holloway）：普羅米修斯號主要科學官之一，伊莉莎白·肖博士的老公，深愛伊莉莎白，為人正直。他在LV-223行星的考察之旅中，因感染Chemical A0-3959X.91 – 15，不幸身亡。

彼得·維蘭德（Peter Weyland）：維蘭德-尤坦尼集團（Weyland-Yutani）的創始人、前最高負責人，也是「普羅米修斯計劃」的贊助人。2091年，維蘭德公司公開宣佈，創始人彼得·維蘭德爵士「逝世」，享年101歲。
